IL-2 R beta & IL-15 R alpha & IL-2 R gamma

Source

Human IL-2 R beta & IL-15 R alpha & IL-2 R gamma Protein, Twin Strep Tag&His Tag (ILG-H5284) is expressed from human 293 cells (HEK293). It contains AA Ala 27 - Asp 239 (IL-2RB) & Ile 31 - Thr 205 (IL-15RA) & Leu 23 - Asn 254 (IL-2RG) (Accession # P14784-1 (IL-2RB) & Q13261-1 (IL-15RA) & P31785-1 (IL-2RG)).

Predicted N-terminus: Ala 27 & Ile 31

Molecular Characterization

The protein has a calculated MW of 29.9 kDa & 52.6 kDa. The protein migrates as 37-39 kDa, 41-43 kDa and 95-120 kDa when calibrated against Star Ribbon Pre-stained Protein Marker under reducing (R) condition (SDS-PAGE) due to glycosylation.

The protein is designed as a heterodimer.

Endotoxin

Less than 0.01 EU per μg by the LAL method / rFC method.

Purity

>95% as determined by SDS-PAGE.

>95% as determined by SEC-MALS.

Formulation

Lyophilized from 0.22 μm filtered solution in PBS, pH7.4 with trehalose as protectant.

バックグランド

Interleukin-2 receptor beta (IL-2Rβ, CD122) and the common gamma chain (γc, CD132) constitute the shared signaling modules for IL-2 and IL-15. IL-15 receptor alpha (IL-15Rα, CD215) is a specific high-affinity binding chain for IL-15. Its sushi domain mediates tight binding, and it can be shed as a soluble form. Unlike classical cytokine receptors, IL-15Rα often presents IL-15 in trans to adjacent cells expressing IL-2Rβ/γc, a process essential for the development, survival, and proliferation of natural killer (NK) cells and memory CD8+ T cells. IL-2 utilizes IL-2Rα, IL-2Rβ, and γc for high-affinity signaling. These three chains are members of the common gamma chain family. γc associates with JAK3, while IL-2Rβ recruits JAK1, both activating the STAT5 pathway to regulate immune responses.
