Betacellulin (BTC)​ is an epithelial growth factor belonging to the epidermal growth factor (EGF) family, primarily known for its potent mitogenic and differentiating effects on pancreatic and other epithelial tissues. In stem cell-based pancreatic differentiation protocols, Betacellulin plays a critical role in the final stages of endocrine maturation. It promotes the functional maturation of pancreatic endocrine precursors, particularly enhancing the differentiation and maturation of pancreatic progenitor cells into glucose-responsive, insulin-secreting beta cells. Additionally, Betacellulin contributes to the expansion and survival of beta-like cells, making it a key component in protocols aimed at generating functional islet organoids or beta-cell clusters for diabetes research and cell therapy development.

バックグランド

Betacellulin (BTC) is a member of the epidermal growth factor family that is comprised of at least ten proteins including EGF, TGF-alpha, amphiregulin, HB-EGF, and the various heregulins. All of these cytokines are synthesized as transmembrane precursors and are characterized by the presence of one or more EGF structural units in their extracellular domain. Betacellulin is expressed predominantly in the human pancreas and induces the differentiation of a pancreatic acinar cell line (AR42J) into insulin-secreting cells, and signals through EGF receptor and ERBB4. It activates ERK and AKT pathways, which induces neural stem cell proliferation and prevents spontaneous differentiation in culture.
