This kit is developed for determining the percentage of human mature lymphocyte subtypes, T cells, B cells, NK cells, T helper/inducer and T suppressor/cytotoxic cell subsets in human whole blood or/and cryopreserved PBMCs by flow cytometry.
It is for research use only.
背景（Background）
PBMCs are a valuable resource in drug development. T cells, B cells, and NK cells are the major lymphocyte subsets,
Flow cytometry has become the technology of choice for the monitoring of immune cells as it provides the possibility to analyze a large number of parameters simultaneously, in a short time and for a reasonable cost.
原理（Assay Principles）
ClinMax™ Human TBNK 6-Color Cocktail Flow Panel used 6 kinds of fluorescent labeled antibody (CD3, CD4, CD8, CD16/CD56, CD19, CD45) premix to detect the proportion of different immune cells in human peripheral blood lymphocyte samples in vitro. This kit uses flow cytometry to analyze the composition and proportion of human mature lymphocyte subtypes, such as T cells (CD3+), B cells (CD19+), T helper/inducer cell subsets (CD3+CD4+), T suppressor/cytotoxic cell subsets (CD3+CD8+) and NK cells (CD3-CD16+CD56+).

Store the unopened kit at 2-8 ℃. Avoid using the kit beyond its expiration date.
Opened kit and reconstituted reagent should be stored at 2-8 °C and the shelf life is 30 days from the date of opening.

TBNK cells gating logical and PBMC cell analysis: Set the CD45/SSC scatter plot to identify the CD45+ lymphocyte cell population (A). CD45+ as lymphocyte cell population, select the CD45+ gate, set the CD3/SSC scatter plot to identify the CD3+ T cell population (B). The CD3/CD4 scatter diagram the Th gate to identify the CD3+CD4+ helper T cell population (C). The CD3/CD8 scatter diagram Tc gate to identify the CD3+CD8+ toxic T cell population (D). The CD3/CD19 scatter diagram B gate to identify the CD3-CD19+ B cell population (E). At last the CD3/CD16+CD56 scatter plot was set to acess the CD3-CD56+CD16+ NK cell population (F).
